穴位埋线对癫痫大鼠认知及PI3K-AKT信号通路的影响Effect of acupoint thread embedding on cognition and PI3K-AKT signaling pathway in epileptic rats

摘要(Abstract):
目的观察穴位埋线对癫痫大鼠认知及PI3K-AKT信号通路的影响。方法 80只SD雄鼠随机分为空白组、模型组、西药组及埋线组,每组20只。模型制造成功后分别进行相应干预共28 d,预定时间行水迷宫实验及处死取海马行TUNNEL、Western Blot法检测PI3K-AKT信号通路相关因子PI3K及Akt的蛋白表达的情况。结果 Morris水迷宫实验:模型组大鼠潜伏期明显增多,空间探索实验中模型组的穿越平台数明显少于空白组(P<0.05);西药组及埋线组大鼠的潜伏期时间较模型组均明显缩短,穿越平台数均明显增多(P<0.05)。TUNNEL染色:模型组大鼠海马TUNNEL染色阳性细胞数目明显增多(P<0.05);西药组及穴位埋线组海马TUNNEL染色阳性细胞数目均较模型组明显减少(P< 0.05)。Western Blot PI3K、Akt蛋白表达:大鼠癫痫发作后模型组、埋线组和西药组的PI3K及Akt蛋白表达均快速上升,经西药及穴位埋线干预后PI3K、Akt蛋白表达均有不同程度的下降,其中以埋线组蛋白表达下降最多。结论穴位埋线可以通过激活癫痫大鼠PI3K-AKT信号通路来抗神经元细胞凋亡,减少海马神经元损伤及改善认知。
